Analysis of temporal retinal sequences

This thesis deals with a method of the retinal image acquisition by fundus camera, their properties and evaluation of the retinal vessel diameter from acquired images. Several studies dealing with usage of retinal vessels diameters for hypertension, cardiovascular diseases, diabetes mellitus and glaucoma are described. The second part of this thesis is focused on the extraction of the retinal blood-vessels pulsation from retinal sequences and the basic processing of extracted pulsation signals. The proposed method is described in details and tested on one dataset.

Author: Mgr. Roman Juřík Title: The effect of modifications in resistance training parameters on blood pressure values during and after the training session. Objective: The aim of this study was to evaluate the acute effect of various forms of agonist- antagonist paired set and agonist resistance training on selected cardiovascular parameters in individuals with normal blood pressure and stage I hypertension. Methods: This research was conducted as a single-blind, controlled study. Based on a systematic literature search, specialized resistance and aerobic training sessions were designed. The program lasted four weeks and included a familiarization phase lasting one week. Prior to the cross-sectional study, anthropometric measurements, biochemical sample collection, muscle strength and fitness testing, and subjective assessment of training intensity using the Borg Scale were conducted. The study consisted of four different resistance training sessions and one aerobic session. Resistance training were categorized into two basic methods: agonist- antagonist paired set and traditional (agonist) training, as well as based on targeted muscle groups (upper and lower body). Molecular mechanisms linking adipose tissue dysfunction and atherosclerotic complications

Obesity and its associated comorbidities increase the risk of mortality. The development of subclinical inflammation associated with obesity also plays a significant role in this. In our work, we focused on changes in the endocrine and mitochondrial function of adipose tissue and their relationship to cardiovascular complications. The first part of the thesis focuses on determining the expression of mitochondrial and endoplasmic reticulum genes in epicardial and subcutaneous adipose tissue, intercostal muscle or right myocardial atrium in patients with atherosclerotic coronary artery disease. Patients with coronary artery disease had reduced expression of several mitochondrial chain genes in epicardial adipose tissue compared to subcutaneous adipose tissue and other tissues, while no change in endoplasmic reticulum gene expression in the above-mentioned tissues has been demonstrated. Based on decreased mitochondrial gene expression in patients with coronary artery disease, we conclude that mitochondrial dysfunction of epicardial adipose tissue may contribute to the development of coronary atherosclerosis. Characterization of epicardial, pericardial, and perivascular adipose tissue in humans and mice

Heart-associated adipose tissue plays an important role in the physiology of the cardiovascular system. Not only does it serve as a protection against mechanical and thermal stress, but it also provides energy for the cardiomyocytes. On the other hand, it can also act as a tissue that promotes a proinflammatory state around the heart, which can result in the development of cardiovascular diseases. Information from the study of epicardial, perivascular or paracardial and other adipose tissue surrounding the heart and large vessels may be used in the prevention of pathological conditions of the heart that are associated with the presence of this adipose tissue.
Validation of the PCR method for the determination of isotype variants in the ApoE gene

Apolipoprotein E (APOE) was discovered in the early 1970s as a 34 kDa polypeptide of 299 amino acids. It plays an important role in the metabolism of cholesterol and triglycerides. It is an important component of VLDL, HDL and chylomicrons. The APOE gene is located on the long arm of chromosome 19 in region 19q13.32. It consists of three introns and four exons. It occurs in three alleles e2, e3 and e4, which encode three different isoforms. The isoforms differ in two amino acids (arginine and cysteine) at positions 112 and 158. The e3 allele is the most common, the rarer alleles e2 and e4 are associated with various types of diseases, such as cardiovascular disease, Alzheimer's disease or type III hyperlipoproteinaemia. The aim of my bachelor thesis was to write a research on the topic using information obtained from available proffesional publications, to master basic methods of molecular biology (which include DNA isolation, DNA concentration measurement, preparation of PCR reaction and gel electrophoresis) and to perform PCR method validation. The theoretical part contains general information about lipoproteins, associated with them lipoprotein metabolism and apolipoproteins. It also includes knowledge of the APOE gene, its product and the effect of APOE on various diseases. Finally, there is an overview of laboratory methods used for genetic analysis of isotype variants in the APOE gene. The practical part, which I did in the genetic laboratory GENLABS s.r.o in České Budějovice, includes the analysis of the APOE genotype and the validation of the PCR method, which was successful. Therefore, the method can be included in the daily operation of the laboratory.
Spatial patterns of links between temperature extremes and cardiovascular mortality in the Czech Republic

Aleš Urban Previous studies have examined relationships of high and low air temperatures to mortality due to cardiovascular diseases (CVDs) in the Czech Republic as a whole. Much less has been understood about possible regional differences in the heat and cold effects on mortality. Within four papers published in international peer-reviewed journals, the author of this thesis investigated links between extreme temperatures and CVD mortality in the Czech Republic while considering in particular differences between (i) urban and rural areas, (ii) regions with different socioeconomic status, and (iii) regions with different physical-environmental conditions. Various biometeorological approaches were compared in order to identify meteorological characteristics affecting heat- and cold-related mortality. Excess mortality was determined as differences between observed and expected daily values, the latter being adjusted for long-term changes, annual and weekly cycles, and epidemics of influenza/acute respiratory infections. Role of perivascular fat tissue in the development of cardiovascular diseases

Abnormal vascular smooth muscle cell (VSMC) proliferation is thought to play an important role in the pathogenesis of atherosclerosis. Adipocytes produce several paracrine bioactive substances that can affect VSMC growth and migration. Our study focused on the ability of epicardial adipocytes to produce bioactive substances together with studying of direct effect of these substances on the VSMC proliferation rate. The gene expression of human cytokines (IL-6, IL-8, IL-18, RANTES and MCP-1) and adipokines (leptin and adiponectin) was measured in primary cell lines of epicardial and visceral adipocytes, both in undifferentiated and mature statuses. Moreover, adipokine production (IL-6, IL-8, MCP-1, VEGF and adiponectin) in conditioned media obtained from above mentioned primary cell cultures of adipocytes was measured by a Luminex assay. The VSMC proliferation rate was measured after co-culturing with CM obtained from primary cell cultures of adipocytes. The epicardial preadipocytes showed an increased expression of IL-8 (3,25-fold, p<0,05) compared with visceral preadipocytes. The expression of the adiponectin in epicardial preadipocytes was markedly decreased in comparison of the expression in visceral preadipocytes (p< 0,0001). Importance of primary and secondary prevention in cardivascular disorder

1 Abstract: Title: The importance of primary and secondary prevention of cardiovascular disease Goals: Develop a theoretical basis for the selected topic. Describe the basic principles of primary and secondary prevention, and thus prepare the groundwork for creating questionnaires. Questionnaire compiled to complement the information obtained from the literature on insight into the practice. Confirm or refute the hypotheses chosen on the basis of a questionnaire. Discuss the most frequently occurring phenomena and compare them with the latest reported information. Draw conclusions and recommendations. Method: For the theoretical treatment of the retrieval method was used. I compared the information and inclined to the best proven. The theoretical part resulted in the goals and tasks of work. For the practical part was conducted by questionnaire survey, which has additional information on the issue of primary and secondary prevention. In the end I obtained data compared with information obtained from studying literature. Results: Lack of information on primary and secondary prevention is mainly for young people aged 10-15 years. Role of the FTO gene in the genetic determination of common multifactorial diseases

Obesity is a risk factor for development of cardiovascular disease, diabetes type 2 and some cancers. Newly detected genetic risk factor for body weight is the FTO gene ("fat mass and obesity associated"). The aim of this thesis was determine 1) whether the presence of risk alleles correlate with BMI in Czech population and to determine 2) whether there is an association between variants in the FTO gene and risk of myocardial infarction/ acute coronary syndrome (MI/ ACS), 3) renal failure (ESRD), or 4) incidence of colorectal cancer (CRC). We analyzed polymorphisms rs17817449 (first intron) and rs17818902 (3rd intron) using by PCR-RFLP and then also RT PCR. We found an association of the first intron variant (but not the 3rd one) and BMI in Czech control population. We have detected an association of 1st intron SNP and BMI changes during the intervention study in obese children, but not in obese females. We found a correlation between the risk allele and increased risk of ACS (OR 1.49) in patients with MI. In patients with ESRD was detected association between the risk allele and the risk of disease (OR 1.37). We didn't confirmed the association between rs17817449 and the development of CRC.
